Glycolysis is an evolutionarily conserved series of ten chemical reactions that utilizes eleven enzymes to concomitantly generate pyruvate and ATP from glucose. Phospho-fructose kinase-2/fructose 2,6-bisphosphatase (PFK-2) stimulates the synthesis and degradation of fructose 2,6-bisphosphate. Pyruvate dehydrogenase (PDH) catalyzes the oxidative decarboxylation of pyruvate to generate acetyl-coenzyme A, thereby linking glycolysis to the TCA cycle with respect to substrate. The phosphorylation of PDH is catalyzed by pyruvate dehydrogenase kinase (PDK), the activity of which is stimulated by the products of PDH catalysis. TIM (Triosephosphate isomerase) exists as a homodimer and catalyzes the reaction of D-glyceraldehyde 3-phosphate into glycerone phosphate.
