How to Get Started
Large Scale Adenovirus Amplification & CsCl Purification
First, we infect ~4000cm2 of 293 cell monolayer through a series of amplifications from customer-provided viral stock or any adenovirus from our catalog. Then, once cytopathic effects begin to manifest, we harvest and lyse the cells to release the virus. Next, the adenovirus is purified via centrifugation using two sequential cesium chloride gradients. The final product is desalted, then titered both spectrophotometrically for viral particles and via plaque formation assay for PFU/IFU. Upon completion, you will receive a final viral stock of approximately 5×1012 to 1×1013 viral particles (~1-3×1011 PFU)*. This process takes ~2-3 weeks to complete.

The process is similar to the large scale amplification. First, we infect ~1000cm2 of HEK293 cell monolayer through a series of amplifications from customer-provided viral stock or any adenovirus from our catalog. Once cytopathic effects begin to manifest, we harvest and lyse the cells to release the virus. Then, the final product is titered for PFU/IFU. Upon completion, you will receive a final viral stock of approximately 2ml with titer of 1-5×1010 PFU/IFU *. This process takes ~2 weeks to complete. * Final viral yield may depend on the transgene in the virus.
