Itch is an HECT-type E3 ubiquitin ligase that promotes the ubiquitination of Notch. The drosophila ortholog of Itch, suppressor of deltex (SU(DX)), was originally identified as a negative regulator of the Notch pathway. Deltex is a transmembrane ligand for Notch and also has a mammalian homolog, DTX1.The human ortholog of Itch and (SU(DX)) is called AIP4 (atrophin-1-interacting protein) and was first described as a protein that interacts with the product of the gene responsible for dentatorubral and pallidoluysian atrophy (DRPLA). In mice, Itch was identified as a mutant locus in 18H mice that resulted in a fatal disease characterized by inflammation of the skin which resulted in constant itching and scarring.

ITCH encodes a member of the Nedd4 family of HECT domain E3 ubiquitin ligases. HECT domain E3 ubiquitin ligases transfer ubiquitin from E2 ubiquitin-conjugating enzymes to protein substrates, thus targeting specific proteins for lysosomal degradation. The encoded protein plays a role in multiple cellular processes including erythroid and lymphoid cell differentiation and the regulation of immune responses. Mutations in this gene are a cause of syndromic multisystem autoimmune disease. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.

About Storage Conditions
All our viral products should be kept at -80°C. At this temperature, the virus will remain stable for 6-12 months (and in some cases, up to 2 years). Once thawed, the product can be stored at 4°C for 2-3 weeks without significant loss of biological activity.
We recommend aliquoting your vectors into low protein binding tubes upon receipt. This helps avoid repeated freeze-thaw cycles, as well as prevent loss of virus. To maintain accurate titer, aliquot in at least 20ul per tube.
